Abstract
Some polymeric constituents of Puccinia graminis tritici urediospores are described. Prolonged extraction of the spores with hot water affords polysaccharide material containing D-mannose, D-galactose, and D-glucose. A levorotatory mannan, separated from this material by copper complexing, is shown to possess a highly ordered structure in which isolated β-1,4- and β-1,3-linked units alternate along the linear chain. The galactosyl units of the extracted polysaccharide are present in the furanosyl form. Acetolysis of the insoluble spore residues shows that pentoses and glucosamine are additional constituent sugars.Ribonucleic acids are isolated by extraction with phenol followed by ion-exchange chromatography, and similar material is obtained from the hot water extract. The phenol treatment also releases much of the spore protein, which is recovered in a highly purified condition.
