MECHANISMS OF RENAL EXCRETION OF CISDICHLORODIAMINE PLATINUM

Abstract
Cis-dichlorodiammine Pt (CDDP) exhibits activity against a wide range of solid tumors and is used in the treatment of epidermoid carcinoma of the head and neck. A major limiting factor in its use has been its nephrotoxicity. Eighteen diuretic rabbits were given CDDP and the clearance of plasma water Pt was determined with a simultaneous inulin clearance. The rabbits were then given probenecid (50 mg/kg) or choline (50 mg/kg) and the clearance of free Pt was calculated. CDDP has a high clearance, exceeding glomerular filtration rate. The drug appears to be secreted by the anionic transport system in the proximal tubule, as the clearance of free Pt is markedly decreased by the administration of probenecid.This publication has 6 references indexed in Scilit:
