Nephelometric Determination of Glycogen in the Rat Liver Glycogen Deposition Assay of Adrenal Steroids

Abstract
A precise, rapid, and simple nephelometric method for rat liver glycogen is described. Livers of rats are digested in hot 30% KOH and diluted to 100 ml. with water. Readings are made on suspensions formed by 2 ml. of the diluted digests and 42 ml. of 57.5% ethanol. Glycogen is quantitatively determined using a standard curve for rat liver glycogen. Livers of uninjected rats are digested, diluted, and pooled as a diluent for the glycogen standard and standardization of the nephelometer. A 98.19% recovery of glycogen from prepared solutions was obtained with a precision of [plus or minus] 0.62%. Comparisons of liver glycogen deposition assays using the method of the National Formulary XI for adrenal cortex injection and nephelometry as described indicate good agreement.Keywords
