Specific Determination of Hydroxy-L-Proline in Biological Materials.

Abstract
A new, specific method for hydroxy-L-proline is reported, based on the development of a characteristic chromagen through ninhydrin development of paper chromatograms followed by rapid treatment with dilute alkali and dilute acid. The new chromagen has an absorption maximum at 550 to 560 m[mu] and fluoresces strongly under UV light. Application of the method to complex biological media showed that 5-10 [mu]g of hydroxyproline can be detected. A high level of this amino acid is shown to occur in chick embryo extract.Keywords
